中文摘要: 金银花有较高药用价值和广泛的应用领域,但是,其植物来源复杂,品种混乱严重,药材质量参差不齐,在生产和交流中缺乏主要有效成分生物合成、积累的生理学机制研究,也缺乏操作性强的质量控制手段,严重制约了金银花的高效、合理利用。为解决这些问题,本课题开展以下研究(1)利用组织化学和免疫细胞化学技术研究金银花花发育过程中主要成分生物合成、转运和积累的细胞学机制研究;(2)运用植物化学、生理学和分子生物学相结合手段研究金银花花器官发育过程中主要成分积累动态及其与调控关键酶活性及相关基因的作用关系;(3)利用ATR-FTIR和LC-MS及HPLC-DAD方法测定不同来源金银花及其易混淆种的化学成分指纹图谱,建立药材质量快速鉴定的数学识别模型。研究结果将从生理生化、亚细胞及分子水平系统阐述金银花主要成分生物合成机制并构建药材质量的快速鉴定体系,为金银花品种改良、合理采收及利用及药材质量控制提供有效技术手段。

英文摘要: Lonicera japonica Thunb. has high medicinal value and widespread application fields, however, its plant source is complicated, variety is confused, and the quality of medicinal material is irregular. Research on physiological mechanism of the biosynthesis and accumulation of main effective components remains limited, and operative methods for quality control is insufficient in production, respectively. An efficient and appropriate utilization of L. japonica is therefore significantly restricted by the issues mentioned above. To address these issues, this project plan to carry out creative researches as follows: (1) Analyze the cytological mechanism of synthesis, transport and accumulation of main components during floral organ development of L. japonica by means of histochemistry and immunocytochemistry; (2) Investigate the dynamic accumulation of main components and its relationship with the expression quantity of key enzyme-encoding genes, also the enzymatic activity downstream during floral organ development of L. japonica using phytochemistry, physiology and molecular biology methods; (3) Detect the fingerprints of chemical composition from Lonicera sources using ATR-FTIR, LC-MS, and HPLC-DAD etc. Meanwhile, a rapid mathematical identification model for quality assessment of Lonicera resources will be built in combination with modern chemometrics. Our results will systematically interpret the biosynthesis mechanisms of main components from physiological, biochemical, subcellular and molecular level. Moreover, a system for quality assessment of Lonicera sources will be built. Ultimately, these achievements will provide efficient techniques for improvement of varieties, rational recovery, and quality control of medicinal materials of Lonicera resources.
