中文摘要: 越来越多的研究认为炎症参与房颤的高凝的发生,但具体机制尚不完全清楚。我们团队在前期的研究中发现在房颤患者外周血中,中性粒细胞产生细胞外DNA和微粒,同时也发现参与炎症的单核细胞也产生微粒,这些微粒及其起源细胞表达磷脂酰丝氨酸。我们在预实验中观察到中性粒细胞释放的细胞外DNA和膜表面外翻的磷脂酰丝氨酸与房颤的凝血有关,我们推测这可能是炎症参与房颤高凝的一个机制。为证实这一假说,我们将入选非瓣膜性房颤患者,采用免疫荧光等技术,观察中性粒细胞细胞外DNA和磷脂酰丝氨酸外翻对于房颤高凝状态的作用及机制。本课题将从新的视角阐明炎症参与房颤高凝的机制,并为房颤血栓栓塞事件的防治提供新的靶点。

英文摘要: More and more studies suggest that inflammation is involved in hypercoagulability of atrial fibrillation (AF), but the precise mechanism remains unclear. Our previous studies have found that neutrophils can release extracellular DNA and microparticles (MPs) into peripheral blood in patients with AF. We also found that monocytes involved in inflammation could produced MPs, these MPs and MP-origin cells expressing phosphatidylserine. In our pre-experiment, we observed extracellular DNA released from neutrophil and phosphatidylserine exposure of neutrophil are associated with coagulation in AF. So we speculate that this may be a mechanism of inflammation is involved in hypercoagulability of AF. To confirm this hypothesis, we further observated the mechanism of extracellular DNA released from neutrophil and phosphatidylserine exposed in the hypercoagulable state of AF using immunofluorescence technique in patients with non-valvular atrial fibrillation. This topic will reveal that inflammation is involved in the mechanism of hypercoagulability in AF from a new perspective, and may provide a new target for the prevention and treatment of AF thromboembolic events.
