中文摘要: Toll样受体(TLRs)是广泛表达于免疫细胞表面的一类模式识别受体,中药的免疫调节作用可能与该信号通路有关。中药多成分、多途径、多靶点的作用特点在免疫调节方面具有广阔前景,然而其复杂性又给物质基础和质量评价研究带来障碍。药效筛选是中药药效物质基础研究的重要环节,其中受体参与机体的各种生理和病理过程,是药物筛选的主要靶标。基于受体的高通量筛选技术在中药有效成分筛选中有特殊优势。本课题以常用药材三七为研究对象,以TLRs受体报告基因技术研究其免疫调节作用的物质基础,并应用于三七药材的质量评价。分别从大、小分子两类物质着手,活性导向的色谱分离手段分段追踪活性成分,通过巨噬细胞和B淋巴细胞进行药效验证以确定有效成分。依分子类型建立相应的分析方法用于三七药材的质量评价。本研究采用高通量筛选法从植物中寻找新的TLRs 激动剂, 有利于推进中药免疫调节功能的研究,并为开发天然免疫促进剂提供新的思路。

英文摘要: Toll-like receptors (TLRs) is a class of pattern recognition receptors which widely express on the surface of immune cells, immune regulation effect of TCMs may be related to the signal pathway. Traditional Chinese medicine (TCMs) has features of multi-component, multi-channel, and multi-target, which has broad prospects in terms of regulating the immune system. However, the complexity of the TCMs make obstacles to the study of material basis and quality evaluation of Chinese medicine. Drug screening is critical part of in the research on pharmacological substance. Receptors are involved in various physiological and pathological processes of the body, and become one of the main targets for drug screening. Receptor-based high-throughput screening technology has its unique advantage in the active ingredient screening in TCMs. Here we take Radix notoginseng, a commonly used Chinese medicine for the study. Searching its immunomodulatory components based on TLRs receptor gene technology material and applies them for the quality evaluation of notoginseng. Respectively, the study will proceed from Macro- and small molecules, the active ingredient are tracked under the guidance of TLRs receptor gene technology in the segments of chromatographic separation and their efficacy are verified by macrophages and B lymphocytes, and then establish appropriate analytical methods for quality assessment in accordance with molecular types. From this study, using high-throughput screening to find new TLRs agonists from plants will help promote the study on immunomodulatory function of Chinese medicine, and provide new ideas for the development of natural immunostimulating agents.
