中文摘要: 光动力疗法是一种基于光敏剂、氧和激发光的微创肿瘤治疗技术,对肿瘤有高度选择性杀伤且基本无毒副作用,其中光敏剂是实现光动力治疗疗效的基础和关键。相较于空心富勒烯分子的光敏特性,金属富勒烯Gd@C82和Sc3N@C80的笼内离域π键体系进一步扩大,并且吸收波长发生红移,从而具有更高的单线态氧产率和组织治疗深度,基于此,本项目拟采用这两种金属富勒烯制备新型光敏剂材料,并系统研究本体结构、衍生化方式、功能化基团的种类和加成数目对癌细胞光诱导损伤的影响,以此为基础,进一步优化金属富勒烯光敏剂材料的设计,最终合成出具有高效杀伤性和生物相容性的光治疗功能材料进行动物活体的光动力实验。本项目将为金属富勒烯用于光动力治疗提供一个切实可行的方案,从而为拓展金属富勒烯的化学反应及实际应用做出有意义的探索。

英文摘要: Photodynamic therapy is a minimally invasive tumor treatment technology based on photosensitizer, oxygen and excitation light with a highly selective destruction and basic non-toxic side effects. Photosensitizers are crucial to photodynamic therapy. Fullerenes can generate large reactive oxygen species upon irradiation with UV or visible light to damage tumor cells. Metallofullerenes,Gd@C82 and Sc3N@C80, with a much more extended π-conjugation and red-shifted optical absorption, facilitate the actual requirements for photodynamic therapy. So in this project, we propose to design and synthesize the novel metallofullerenes-based photosensitizers for photodynamic therapy. We will derivatize the two with different reactions to obtain three categories water-soluble compounds. Then, study how the frame stuctures, derivatization methods and numbers and types of functional groups influence the results of photo-induced cytotoxicity against in vitro tumor cells,and finally choose and further optimize the high-efficient ones to prove their photodynamic effect on mouse tumor in vivo finally. Our project will not only provide an available solution for endohedral fullerenes used as photosensitizers, but also contribute significantly to the chemical derivatization and practical applications of endohedral fullerenes.
