中文摘要: 新疆Bt棉花的大面积种植有效控制了靶标害虫棉铃虫,但也带来了棉铃虫对Bt棉花产生抗性的潜在风险。本项目基于近年来国内外对Bt抗性的研究进展、新疆Bt棉花种植分布现状和前期抗性监测基础,利用F1/F2代法监测新疆不同地区棉铃虫种群对Bt棉花的耐受性,评估棉铃虫对Bt耐受性的地区间差异;调查不同地区各种寄主作物的种植比例和各代次棉铃虫种群的发生密度,评价棉铃虫的寄主来源和不同寄主作物的庇护所功能;通过主要蛋白酶和解毒酶酶活测定,解析不同地区棉铃虫中肠酶活性差异与其耐受性的相关性;采用DNA分子技术筛选棉铃虫不同地理种群抗性基因,明确新疆棉铃虫种群产生耐受性的关键基因。通过本项目研究,对新疆不同地区棉铃虫种群的Bt耐受性进行评价,从生态、生化和分子多重角度解析棉铃虫自然种群产生耐受性的影响因素,探索出适于新疆棉铃虫种群耐受性的监测技术,为制定新疆Bt棉花靶标害虫抗性综合治理策略提供理论依据。

英文摘要: Cotton bollworm [CBW, Helicoverpa armigera (Hübner)] has been effectively suppressed by area-wide Bt cotton adoption in Xinjiang, but has the potential to develop the resistance to Bt cotton. Based on the relative studies on Bt resistance of CBW worldwide and the status of Bt cotton planting in Xinjiang, at 15-20 cotton planting areas in Xinjiang, we will monitor CBW tolerance to Bt cotton using F1/F2 method, and evaluate the function of different host crops as the natural refuge of CBW by investigating the planting rate of different crops and the density of CBW in each crop per generation, and measure the activity of proteases and detoxifying enzymes in the CBW midgut, then explore the major genes relative to Bt resistance of CBW. By above-mentioned studies, we will evaluate CBW tolerance of different geographical populations, and analyze the major factors resulting in different tolerance to Bt cotton, then develop biochemical and molecular techniques for monitoring CBW tolerance in Xinjiang. All these are available for developing the strategy of Bt resistance management of CBW in Xinjiang.
