中文摘要: 耳蜗的高听觉灵敏度来源于科蒂氏器内部复杂的生物力学和电化学等过程,总称为耳蜗放大器(CA)。CA的空间分布以及产生机制一直是耳蜗动力学领域的研究热点,但是至今没有完全理解。本研究将:1)运用先进的近红外低相干干涉仪系统,在体测量网状板(RL)和基底膜(BM)的振动频谱在耳蜗长度方向的分布,并由此计算出外毛细胞(OHC)伸长缩短运动频谱的分布,并以此确定CA在耳蜗长度方向的位置和分布。2)利用RL和BM空间分布测量结果,结合数学模型对科蒂氏器内部沿长度方向的耦合机制进行研究,理解它们对CA产生的影响。本研究的意义在于采用全新的光学方法对科蒂氏器内部结构振动的空间分布进行扫描测量,并提出了以OHC伸长缩短而不是传统的BM振动来估计CA。并以此扫描数据结合模型对耳蜗长度方向耦合机制进行了研究,对CA产生机制的提供了新视角。

英文摘要: The high hearing sensitivity of the mammalian cochlea results from a complex biomechanical electro-chemical process, called cochlear amplifier (CA). The location and the spatial distribution, as well as the CA generation mechanism have been hot topics in the field of cochlear mechanics but remained unknown until now. In this study, we will: 1) Measure the longitudinal distribution of the RL and BM vibration spectra in vivo, and calculate the OHC length change so as to estimate the location and longitudinal distribution of the CA. 2) Based on the spatial distribution of RL and BM vibration data, study the longitudinal coupling mechanisms inside the organ of Corti with a mathematical model so as to understand its contribution to the CA generation. The significances of this study are 1) The in vivo measurement of vibration spectra of internal structure of organ of Corti using low-coherence interferometry technique; 2)The method to estimate the CA spatial distribution from the OHC activity, rather than the traditional BM vibration spectra.With the combination of novel experimental data with modeling effort, this study will provide a new angle for understanding CA generation.
