中文摘要: 针对膜蛋白质组学中膜蛋白质混合物系分离纯化需求,研究电场强化作用下双水相逆流色谱机制及对疏水性膜蛋白质的分离特性,以期获得高分辨率和高通量的目标膜蛋白质。项目主要内容有:(1)研究表面活性剂双水相体系电场强化下分相特征,确定具有快速分相性能的双水相溶剂体系和外加电场特征参数范围。(2)研究双水相体系在电场强化作用下逆流色谱中相分布特征,提出高固定相保留率的逆流色谱操作参数和电场特征参数指标。(3)研究模型蛋白质在电场强化作用下双水相逆流色谱中的分离特性,探索蛋白质逆流色谱分配过程中理论板模型的各项参数指标及变化规律,用于电场强化作用逆流色谱仪操作条件控制和结构参数设计。(4)分离纯化实际膜蛋白质混合物系中目标膜蛋白质,分析评价电场强化作用双水相逆流色谱对疏水性膜蛋白质混合物系的应用特性。项目的研究对电场强化作用下双水相逆流色谱技术的深入开发及疏水性膜蛋白质混合物系的分离纯化具有重要意义。

英文摘要: In order to satisfy the requirement of membrane proteins separation in membrane proteomics, electrokinetic counter-current chromatography using aqueous two-phase surfactant system for hydrophobic membrane proteins separation are studied so as to achieve the target membrane protein with high resolution and throughput. The following issues are mainly concerned in the program: (1) Investigate the electrokinetic phase separation behavior of aqueous two-phase surfactant system for counter-current chromatography and then determine the type of aqueous two-phase surfactant system and corresponding electric fields with appropriate property ranges. (2) Discuss the phase distribution characteristics of aqueous two-phase surfactant system in electrokinetic counter-current chromatography, and provide operation parameters and electric fields parameters with high stationary phase retention rate. (3) Research on the model proteins separation in electrokinetic counter-current chromatography, obtain the parameters and variation of chromatographic process theoretical plate model for protein separation in electrokinetic counter-current chromatography, and then direct to improve operating condition control and structural parameters design of electrokinetic counter-current chromatography. (4) Separate the target membrane protein in complicated natural mixture, analyze and assess the application performance of electrokinetic counter-current chromatography using aqueous two-phase surfactant system for membrane proteins separation. The program will be of great importance in the further development of electrokinetic counter-current chromatography and the separation of membrane protein mixtures.
