中文摘要: 圆头蒿广布于我国西北部地区,资源丰富,圆头蒿多糖(ASP)的结构未见国内外文献报道,对其研究不仅具有一定的理论意义,更具良好的应用前景。项目组前期进行了ASP的提取纯化,确定了单糖组成和分子量,进行了体内外抗肿瘤活性的初步研究。在此基础上,针对ASP缺乏各层次、深入的结构研究,硒多糖优良的活性及合成过程中结构不明确、硒含量低等问题,本项目拟系统研究ASP的化学结构(单糖组成、糖苷键类型、连接顺序和分支点等);合成不同硒含量的含硒衍生物(SeASP),确定硒含量及存在形式、价态和取代位置等;通过对ASP和SeASP溶液构象的表征,阐明多糖化学结构的变化对溶液构象及转变方式的影响;进行体内外抗肿瘤实验,探讨ASP和SeASP的结构、溶液构象对其抗肿瘤活性应答效应的影响规律;研究凋亡相关基因表达的变化,确定ASP和SeASP诱导肿瘤细胞凋亡的途径及机理,为天然高分子及衍生物的合成和设计提供依据。

英文摘要: Artemisia sphaerocephala is widely distributed in western China. There are no similar reports of the structure studies on Artemisia sphaerocephala polysaccharide (ASP).These are topics of theoretical interest and also of great relevance to applications. It is reported that Se plays an important role in increasing the antitumor activity of Se-containing polysaccharides.In our previous studies, ASP was extracted and purified, monosaccharide composition (molar ratio), molecular weight and antitumor activities were determined. The application of ASP was limited due to the lack of further studies on chemical structure. In this project, chemical and spectroscopic methods were employed for structure characterization (monosaccharide composition, molecular weight, linkage of glycosidic bond and branch point) of ASP. Se-polysaccharides with high Se contents were synthesized. The existing form and valence were characterized. The relationship between primary structure and solution conformation was also studied. Furthermore, anti-tumor activities of both ASP and SeASP were evaluated in vitro and vivo for a better understanding of the relationship between structure and biological response effects. For the purpose of explaining the mechanism of apoptosis, apoptosis related gene expressions were also detected.This study could provide evidence for design and synthesis of biopolymer and its derivatives.
