中文摘要: 早产儿视网膜病变(ROP)是儿童致盲的主要原因之一。最新研究表明某些细胞外循环microRNA特异性调控新生血管的形成,但microRNA与ROP发生发展之间的关系,目前未见报道。基于我们原有的工作基础以及最新的研究结果,结合新华医院的医疗优势,本课题旨在探究细胞外循环microRNA在ROP患者中的特异性表达。拟将ROP和对照组患儿的血浆和玻璃体液,用qPCR array技术筛选出差异表达的microRNA,通过real-time PCR进行验证和分析其与ROP病变程度的相关性;并对筛选出的microRNA通过细胞实验和动物实验对其功能进行研究。课题将为预测ROP病程的进展提供分子靶点,从microRNA角度为ROP发病机制的研究提供新的线索。

英文摘要: Retinopathy of prematurity is one of the major cause of children blindness. New research shows that the circulating microRNA specific regulates angiogenesis. However, the relationship between microRNA and ROP has never been reported. Based on our original work and the new research results, combined with medical advantages of our hospital, the purpose of this study is to explore the expression of circulating microRNA in ROP patients. ROP and control patients plasma and vitreous are going been screened the differential expression of microRNA by using qPCR array technology, and will be verified and analyzed the correlation with the severity of ROP by real-time PCR. The screened microRNA will be studied on its function via cell and animal experiment. This study will provide the molecular target for prediction of ROP, and provide a new clue of the pathogenesis of ROP from the angle of microRNA.
