中文摘要: 药用植物道地性的形成主要是基因型和环境相互作用的结果,而表观遗传在生物应答环境的过程中起到重要作用。DNA甲基化是表观遗传最重要的现象之一,已成为当前研究的热点,但是关于药用植物DNA甲基化的研究比较少,尤其怀地黄DNA甲基化的相关研究尚未见报道。本项目拟以怀地黄85-5的块根为实验材料,应用表观遗传学、结构生物学、植物化学等手段,对栽培于不同产区的怀地黄块根的梓醇积累、基因组DNA甲基化修饰等进行研究,同时应用DNA甲基化抑制剂5-氮杂胞苷胁迫处理的怀地黄块根作对比实验,分析DNA甲基化在怀地黄块根的有效成分积累以及应答不同生态环境中的调节作用,以期揭示DNA甲基化在怀地黄道地性形成过程中的作用,也为从表观遗传学角度研究药用植物道地性的机理提供借鉴和参考。

英文摘要: Geoherbalism formation of medicinal plant is mainly related to the interaction between genotype and environment, while epigenetics plays an important role in organisms adapting to environment. DNA methylation is one of the most important phenomena in epigenetics and has been a hotspot recently, but research about DNA methylation in medicinal plant is less, especially has not been reported in Rehmannia glutinosa f. Hueichingensis Hsiao. In this study, root tuber of Rehmannia glutinosa f. Hueichingensis Hsiao would be experimental material, catalpol accumulation and genome DNA methylation of root tuber from Rehmannia glutinosa f. Hueichingensis Hsiao in different producing areas would be studied by epigenetics, structural biology, phytochemistry and other methods, simultaneously root tuber of Rehmannia glutinosa f. Hueichingensis Hsiao treated with 5-azaC would be contrast experiment, and the correlation among environment, DNA methylation and effective constituent of Rehmannia glutinosa f. Hueichingensis Hsiao would be also analyzed to discuss role of DNA methylation modification in geoherbalism formation of Rehmannia glutinosa f. Hueichingensis Hsiao, which would provide reference to reveal mechanism on geoherbalism of medicinal plants from the perspective of epigentics.
