中文摘要: 肝癌已上升为我国第2位癌症杀手,在世界范围内肝癌的发病率呈明显的上升趋势。因此,尽早诊断与及时治疗是改善肝癌患者预后最有效的手段。目前肝癌的定性诊断依赖血清标志物检测,其中高尔基体蛋白73 (GP73)最有可能成为更好的诊断肝癌,尤其是早期肝癌的血清标志物。本项目以肝癌的早期诊断为目标,以肝癌新型标志物为研究对象,以核酸适配体作为识别单元,基于分子识别和信号传导相分离的原理,结合功能化纳米材料,设计构建信号放大的光电化学核酸传感平台,提出对肝癌新标志物循环放大检测方法,实现对肝癌标志物的敏感性和特异性检测,并进一步分析其与肝癌发病机制的相关性,促进肝癌新型标志物检测方法的发展,为肝癌早期诊断和预防提供参考。

英文摘要: Liver cancer has risen to the second cancer killer in China,the incidence rate of liver cancer shows an obvious rising trend in the world. Therefore, early diagnosis and timely treatment are the most effective means for improving the prognosis of HCC. Today, the qualitative diagnosis of liver cancer serum depend on marker detection, in which the golgi protein 73 (GP73) is most likely to become a better serum markers for diagnosis of liver cancer, especially for the early HCC. Based on the separation principle of the molecular recognition element and signal reporter, this project combines with functional nano-materials to design and construct a photoelectronchemical aptasensing platform with signal amplification, using prevention and treatment of early liver cancer as breakthrough and new marker of liver cancer as research object. Furthermore, the method of cycle amplification detection to new marker is put forward, realizing sensitive and specific detection the hepaticarcinoma marker. Finally, we further analyze the correlation between marker and pathogenesis of hepatocarcinoma. The project will promote the development of detection method for new markers of hepatocarcinoma and provide the reference for the early diagnosis and prevention of liver cancer.
