中文摘要: 目前,甲壳素和壳聚糖基水凝胶在骨修复领域的应用尚处在初期发展阶段,其力学和骨细胞粘附性能有待于进一步提高。本项目拟采用原位聚合丙烯酸法构建高强度双网络结构甲壳素基复合水凝胶,通过其负电性聚丙烯酸预先固定钙离子作为反应位点实现羟基磷灰石粒子均匀地原位合成,系统研究甲壳素水凝胶孔尺寸和聚合条件对其结构和力学性能的影响,揭示羟基磷灰石对其力学和骨细胞粘附性能优化的作用,并阐明相关作用机制。在此基础上,对甲壳素基复合水凝胶进行脱乙酰化处理制备正电性壳聚糖基聚电解质复合水凝胶,深入探究脱乙酰化条件对其组分间相互作用以及结构和性能的影响。本研究工作旨在实现高强度生物相容性甲壳素(壳聚糖)基骨修复材料制备,解决原位合成过程中羟基磷灰石呈梯度分布这一基础科学问题,同时为构筑聚电解质复合水凝胶提供一种新方法和思路,具有重要的科学意义和实用价值。

英文摘要: Recently, the application of chitin and chitosan based hydrogels in the bone repairing field is still in the early developing stage, their mechanical and osteocyte adhesion properties are still needed to be improved. In this project, in-situ polymerization of acrylic acid will be adopted to construct high strength chitin based composite hydrogels with double network structure, whose electronegative polyacrylic acid will be used to fix calcium ions as the reaction site for the homogenously in situ synthesis of hydroxyapatite particles. The effect of the pore size of chitin hydrogels and polymerization conditions on the structure and mechanical property will be systematically studied, the role of hydroxyapatite on the improvement of the mechanical and osteocyte adhesion properties will be revealed, and the corresponding mechanism will be clarified. On this basis, the chitin based composite hydrogels could be converted into electropositive chitosan based polyelectrolyte composite hydrogels through the deacetylation process, the effect of the deacetylation conditions on the interaction between the components, structure and property will be deeply studied. The work aims to realize the preparation of high strength and biocompatible chitin (chitosan) based bone repairing materials, solve the gradient distribution problem for the direct in situ synthesis of hydroxyapatite particles and provide a novel method and thought for the construction of polyelectrolyte composite hydrogels at the same time, showing important scientific significance and practical value.
