中文摘要: 组织工程膀胱的构建取得了阶段性的成果,但目前仍难以大规模的临床应用。阻碍其应用的因素之一是对促进组织工程功能化的物理及化学调节因素知之甚少。国内外及我们的研究发现应力刺激在膀胱发育和疾病发生中起着极为重要的作用。尽管如此,膀胱种子细胞如何对应力做出生物应答反应尚未完全明晰,从而达到细胞功能成熟化。研究显示,血管平滑肌及气道平滑肌细胞在适当的生物力学刺激下,可检测到炎性基因表达升高。同时,众多研究显示M受体在机械应力引起膀胱种子细胞的适应性变化的过程中起到了关键作用。基于应力下膀胱重塑过程,加之炎症在其中发挥重要作用,结合我们前期研究基础,有理由推测M受体在膀胱重塑过程中调控炎症因子的表达,从而调节膀胱功能。本研究拟运用Biodynamic应力加载培养系统,观察应力下各类炎性因子变化及其对膀胱细胞功能的调控,并进一步阐明M受体在膀胱应力生物学转导机中定位及作用。

英文摘要: Although tissue engineering evoloving fast, it is still hard to apply in clinical in large scale.One of barrier is we do not fully understand how the physical and chemical factors to improve the function of tissue engineering.The international combined our study find that mechanical stimulation is significant factors for development and diseases of baldder. However, how bladder cell reponsond to mechanical stimulation is still not clear. Studies find that the inflammatory factors can be upregulated in vascular SMC and air way SMC under optimal mechanical stimulation. At the same time, many studies demonstrate that M receptors are key factors in bladder cell under mechanical stimulation. One of hypothesis in our study derived from our previous research is that the mechanical stimulation regulate inflammatory factors in bladder cells through M receptors, based on the processof bladder regeneration, and inflammatory factors play important role in this biological process. The Bildynamic bioreactor will be applied in our study, the change of inflammtory factors and its role in bladder function will be observed. In addition, the location and role of M receptor in bladder cell under mechanical stimulation will be examined.
