中文摘要: Wnt10b是 Wnt 家族的重要成员之一,具有最明显的限制性表达,对毛发基板的形成和维持非常重要。本课题组在研究中发现:给内蒙古绒山羊在体埋植褪黑素可提早 2 个月诱发新绒生长,提高羊绒产量,经qRT-PCR检测埋植褪黑素上调了皮肤毛囊休止期和退行期Wnt10b基因的表达,可推测Wnt10b参与了褪黑素促进绒毛生长的过程,但其信号通路及调控机制尚不清楚,也鲜有报道。因此,本课题拟采用转录组测序、原位杂交、qRT-PCR、免疫组织化学、Western blot、腺病毒介导Wnt10b基因过表达、RNAi等技术,从体内、体外两方面研究Wnt10b基因在褪黑素促进绒毛生长周期中的表达和作用机制,并利用干扰技术阻断经典 Wnt 信号通路关键因子,寻找Wnt10b 诱导毛囊生长的信号通路,对实现人工调控绒毛生长时间和提高内蒙古绒山羊绒毛产量具有重要指导意义。

英文摘要: Wnt10b is one of the most important members of Wnt family, of which expression is obviously restricted and important to hair plate formation and maintenance. Our prevoius reseach has discovered that implanting melatonin (MT) to Inner Mongolia Cashmere goats advanced the growth of cashmere by two months and improved the quantity of cashmere. The result of qRT-PCR showed, during telogen and catagen phase, the expression of Wnt10b was increased in the Cashmere goats hair follicles after MT implanting, which means that Wnt10b was involved in the process of Cashmere goat hair growth promoted by MT. However, its signal pathway and the regulation mechanism are not clear, and there are few reports on them. Therefore, in this study, the effect and mechanism of MT in promoting the growth cycle of cashmere are to be detected in vivo and vitro, using In Situ Hybridization, qRT-PCR, IHC, Western Blot, Adenovirus mediated purpose gene and RNAi techneques. Furthermore, the signal pathway of Wnt10b inducing hair follicle growth is to be explored by blocking the key molecules in classic Wnt signal pathway with interference technology. The results of this project is significant for revealing the time for artificial regulation of cashmere growth and increasing the production of cashmere of Inner Mongolia Cashmere goats.
