中文摘要: 如何提高乳腺癌的早期诊断和治疗是国内外肿瘤研究领域面临的一个挑战性问题,而基于分子影像的靶向诊断和治疗为乳腺肿瘤的治疗带来新的希望。新近研究表明肿瘤相关巨噬细胞(TAMs)在肿瘤发展过程中发挥着重要作用,如能实现针对乳腺TAMs的早期观测和治疗将会为乳腺癌的早期诊断和有效治疗带来帮助。本项目拟通过将近红外染料IRDye-800CW、靶向TAMs肽片段(M2Pep)和消除TAMs表达的化疗药物曲贝替定结合于载体系统,构建靶向TAMs的诊疗一体化分子探针,结合光学和光声分子成像技术,实现对TAMs在乳腺肿瘤发展过程中的分布进行在体动态监测,探索TAMs对乳腺癌发展的影响及作用,以及对TAMs进行靶向治疗和疗效评估。通过本项目预临床研究将为临床上实现乳腺癌的早期诊断和合理及高效用药提供新的参考依据。

英文摘要: Exploring new adjuvant diagnostic and treatment strategies for the breast cancer is urgently needed. The targeted drug delivery holds the potential for the breast tumor treatment. Recent studies revealed that tumor associated macrophages (TAMs) represent a predominant component of the tumor mass in breast carcinoma, and appear to be an important orchestrator. TAMs are a novel candidate for targeted cancer therapy and prognostic biomarker of response and clinical benefit. In this project, we plan to conjugate PLA-PEG nanoparticles with near infrared dye (NIR) IRDye-800CW, TAMs targeting peptide M2Pep, and Trabectedin (a new chemotherapy drug with effects on decreasing TAMs expression) to develop a theranostic probe. With the aid of optical and photoacoustic imaging, the probe can facilitate the dynamic monitoring of TAMs during the breast tumor initiation and progression, investigating the roles of TAMs on regulating breast tumor development, and evaluating the therapeutic effects of our developed theranostic probe on breast tumor. Our project will provide new preclinical evidence for the early diagnosis and drug therapy of breast cancer.
