中文摘要: 对虾养殖是我国沿海地区重要的产业,而由细菌和病毒等导致的对虾病使该养殖业蒙受巨大损失。目前对虾感染性疾病控制的方法尚不能满足生产需求。清道夫受体(SRs)是一种重要的模式识别受体,在先天免疫中发挥关键作用,具有吞噬、清除凋亡细胞和病原微生物等作用,然而迄今为止,清道夫受体在对虾中鲜有报道。本项目拟对在对虾转录组分析的基础上得到的多种SRs进行性质与功能研究:包括鉴定对虾中存在的不同类型的SRs;分析它们的组织分布和病原感染后的表达模式;利用重组表达或天然的SRs研究其所结合的体内配体和病原微生物来源的体外配体;揭示SRs相互作用的蛋白以及这种相互作用与病原清除和吞噬的关系,阐明SRs与配体结合后所激活的免疫相关信号途径和调控的效应作用及效应分子。该研究在理论上可以丰富对虾先天免疫的理论,在实践上可以为对虾养殖中的疾病防治提供新靶标和新思路。

英文摘要: Shrimp are the major aquaculture species generating higher revenue in costal countries,including China. However, diseases are major impacts on the successful production of these cultured species, especially bacterial and viral diseases. Understanding the shrimp defense system will contribute in control of shrimp diseases. Scavenger receptors (SRs) are the pattern recognition receptors and play important functions in pagocytosis and removal of apoptotic cells and infected pathogens. Almost no reportes about SR functions in shrimp till now. In this research, we will study the functions of shrimp SRs obtained from reancriptome analysis, including the identification of different types of SRs, analysis of temporal and spatial expression, studies of the in vivo and in vitro ligands of different SRs with recombinant or native SRs, the interactions of SRs with other proteins and relationship with removal and phagocytosis of pathogens, clarification of the related signal pathway activation after SRs binding to ligands and the effectors regulated by the signal pathways. These studies will enrich the theory of innate immunity in shrimp and provide new targets or methods for disease control in shrimp aquaculture.
