中文摘要: 林木生长发育过程中根系主要起两方面的作用,即固定和吸收水分及矿质营养。侧根的形成是根系形态建成的一个主要决定性因素,生长素在其中起到关键作用。在前期研究中,申请者发现了PtoWOX11/12a受生长素的诱导且负调控侧根的形成,但调控机理尚不明确。基于生长素在侧根形成中的重要性,申请者提出:PtoWOX11/12a通过与生长素组成负反馈回路调控杨树侧根的形成。本项目首先结合细胞生物学的方法,分析PtoWOX11/12a参与侧根形成的具体阶段;然后确定PtoWOX11/12a对侧根的形成中的功能和对生长素响应的影响;最后探索PtoWOX11/12a与生长素途径之间的相互关系,揭示PtoWOX11/12a负调控生长素响应的分子机制,为充分揭示林木根系形成的分子机理以及基因工程育种提供科学依据。

英文摘要: The root system of tree is responsible for anchorage to the soil as well as for minerals and water uptake. Auxin play crucial role on lateral root formation that is very important for root systems architecture. We found that PtoWOX11/12a that is induced by auxin negatively regulate lateral root formation, however, the molecular mechanism is unclear. So, we proposed that PtoWOX11/12a regulate lateral root formation via negative feedback circuit-mediated auxin response in Populus. Firstly, this project is intended to analysis precise expression pattern of PtoWOX11/12a during lateral root formation process using cell biology method. And then, the function of PtoWOX11/12a and auxin response on lateral root formation will be identified. At last, the interaction between auxin and PtoWOX11/12a will be analysised by RT-PCR and Chip-Seq. It is very important for roor system formation and Populus breeding to uncover the molecular mechanism of PtoWOX11/12a regulaed lateral root formatiom.
