中文摘要: 目前小分子环境污染物在我国食品中残留严重,并对人类健康造成严重威胁,因此建立此类污染物的快速检测方法具有重要社会和科学意义。本项目针对目前电化学生物传感器研究中遇到的检测速度及检测准确性等瓶颈,拟结合免疫分析的高特异性和纳米微粒的增敏特性,建立基于纳米免疫分散电极的电化学生物传感检测方法,并应用于食品实际样品检测。本项目以广受关注的内分泌干扰素双酚A为目标物,通过分子模拟等方法,定向制备高特异性高灵敏度双酚A单克隆抗体,揭示其制备和筛选规律。利用抗体修饰的纳米金磁珠作为分散电极,通过增加传感器表面识别元件承载量,简化电极修饰步骤等途径,达到缩短检测时间和提高灵敏度的双重目的。对不同食品基质中双酚A残留量检测应用进行研究,建立能够应用于多种实际食品样品的快速检测方法,形成相应产品,为我国在小分子环境污染物快速检测技术领域产品自有化提供示范。

英文摘要: The small molecule environmental contaminants in foods which have fatal threat to human health have become more serious in China now. Therefore, it is important to develop the detection methods for monitoring these contaminants. The main aspects of electrochemical biosensors which need to improve include the detection time, the sensitivity, selectivity and reproducibility. To solve these, ours project aims to develop dispersible electrodes based biosensor which combines the high selectivity of immunoassay and the high conductivity and surface-volume ratio of nanoparticles, for real sample detection. With bisphenol-A as the analyte, we aim to generate anti-bisphenol-A polyclonal antibodies via molecule-modeling, and then investigate the principle of the generation. After that, the antibody modified gold-coated magnetic nanoparticles will be utilized as dispersible electrodes in the biosensor fabrication. The method could be time-efficient and highly sensitive, since the biosensor has more receptors and simpler electrode modification steps over the reported ones. The matrix effects will be then studied for the practical use. In the end, the related products will be produced which may spirit the manufactory of the rapid detection products in China.
