中文摘要: 色谱分离是生物下游工程的重要技术,广泛应用于高附加值生物产品的分离和富集,而色谱技术的核心就是色谱介质。因此,研制高效色谱介质及快速分离技术一直是生物化工领域的研究热点。本项目以开发生物快速分离技术为目标,以纤维素基整体柱为研究对象,按照生物色谱材料的使用要求,系统研究纤维素基整体柱制备和应用的基本理论和技术问题。本研究拟用乳化过程理性调控整体柱的多孔结构,揭示乳化致孔机理,进而指导整体柱的结构设计和合成;考察纤维素基整体柱的孔结构特性和色谱性能,探究它们之间的构效关系,明确大孔结构对色谱性能的增强作用;利用模型生物分子,研究整体柱的分离性能和色谱机制,创建实用高效的分离方法;选择实际分离体系评价纤维素基整体柱的分离效果。本研究的意义在于,将材料科学的最新研究成果加以改进后,有效运用到生物色谱技术的前沿领域,不仅可以促进色谱理论和技术的发展,也为纤维素功能材料的绿色合成提供新思路。

英文摘要: Porous monolith has attracted great attention in these days due to its potential application in the separation of large molecules. The popularity lies in its unique bimodal porous structure because the large pores can facilitate mass transport and mesopores can provide abundant binding sites for high adsorption capacity. In this context, a novel cellulose based porous monolith is studied with respects of fabrication, characterization and chromatography application. The research focuses on the porous construction of cellulose based porous monolith, which is realized by the emulsifying process. The pore formation mechanism is then clarified by exploring the effect of emulsifying conditions on the pore generation of cellulose based monolith. After that, the cellulose based monolith is fabricated and characterized according to the separation requirement. Then, more works are processed to analyze and evaluate the cellulose based monolith in terms of porous structure, physical properties, and the separation behavior. Based on these results, the structure-activity relationship is elucidated together with chromatography mechanism. Finally, the cellulose based monolith is modified by suitable ligands and explored carefully toward the actual separation application.
