中文摘要: 利用光照下可产生光敏化作用的光敏剂来破坏病变组织的微创疗法-光动力疗法,因其具有高效、安全、创伤小、副作用少、可协同性、可重复性和相对成本低等优点,而受到了广泛关注。光敏剂是光动力学疗法的关键因素。然而,光敏剂的药物传送效率低,光敏剂光动力效果有限,组织穿透深度浅等问题还有待解决。本项目拟通过采用位于近红外荧光发射波段的光敏剂与金纳米笼材料相结合,有望提高光敏剂的药物传送效率,组织穿透深度和光动力效果。此外,本项目在光敏剂和金纳米笼结合体构建的基础上,制备一种新型的热敏脂质体将其与靶向化疗药物相交联,在近红外激光照射条件下,利用结合体产生的光热效应,使得脂质体达到相转变温度,药物得以释放,最终完成细胞的光热效应、光动力效应以及化疗的协同治疗。而且基于光敏剂的荧光特性,在治疗的同时可进行动态实时无创的可视化监测,实现诊疗一体化的目的。为肿瘤的诊断和治疗提供新方法。

英文摘要: Minimally invasive photodynamic therapy, destroying lesions with light-activated photosensitizer, has been brought into focus for its high efficiency, safety, small operation wound, little side effect, synergy compatibility, repeatability, relatively low cost and so on. Photosensitizer is the core of photodynamic therapy. However, low delivery efficiency and limited photodynamic effect of photosensitizer and shallow tissue penetration depth remain to be solved. In order to enhance the delivery efficiency, photodynamic effect and tissue penetration depth , we combine near-infrared-emissive photosensitizer with gold nanocage. Besides, we will prepare a novel thermo-sensitive liposome conjugated with targeted chemotherapeutic agents on the basis of the photosensitizer-conjugated gold nanocage. With photo-thermal effect induced by near infrared laser, liposomes reaches phase transition temperature and chemotherapeutic agents are released. Hence the synergy of photo-thermal effect, photodynamic effect and chemotherapy is achieved. Furthermore, real-time non-invasive dynamic visualization imaging method based on the fluorescence property of photosensitizer can be conducted during the treatment, which provides a new type of application for tumor theranotics and diagnostics.
